For the Thursday night game against Valencia, Isco was originally included in the squad list of Real Madrid but he was ruled out by a muscle injury.
The midfielder was injured to his right thigh, the club announced on Thursday morning.
He is expected to miss about three weeks of action, which could be six matches.
